A Note on the Production of Acid by Pneumococci. by Glenn

چکیده

The latent period of growth or "lag," first recognized by Mfiller in 1895,1 is the time interval between the inoculation of a medium and the beginning of growth at the max imum rate. Chesney concluded that the latent period in the growth of bacteria is an expression of injury which the bacterial cell has sustained from its previous environment3 This conclusion was based upon the observations that the latent period was evident only when the cultures used in inoculation were no longer growing at the maximum rate, and that pneumococci cultures at the end of the period of maximum growth contain a substance that inhibits the growth of actively growing pneumococci. In studying the nature of the influence which the products of metabolism exert upon the further activity of a microorganism it is important to investigate the changes which may have occurred in the hydrogen ion concentration of the medium. The question of the influence of hydrogen ion concentration has been reviewed exhaustively by S/~rensen, ~ by Michaelis, 4 and more recently by Clark and Lubs3 Clark and Lubs emphasize its importance as one of the factors controlling bacterial growth thus: "Just as we pay attention to the temperature of the incubator, so should we pay attention to the hydrogen ion concentration of the medium." We therefore undertook to study the hydrogen ion concentration of actively growing pneumococcus cultures. 6

A Note on the Production of Acid by Pneumococci

1. Actively growing pneumococci produce acid at such a rate that change in the pH of the medium parallels change in rate of growth. 2. The death of the pneumococci is not followed by change in the reaction. 3. Acidification during growth in beef infusion media proceeds until a pH of about 7 is reached. At this point growth stops. The increase in hydrogen ion concentration is not the only origin...

A Study on Plant Polymer: Pectin Production and Modification (RESEARCH NOTE)

Abstract Pectin from two different plant sources: apple and sugar beet was studied. Apple pectinwas modified by chemical treatments such as: acid, alkali and ammonia, so as to obtain an ester levelequal to or less than 50 %, which is described as low methoxyl (LM) pectin. LM pectin form gelswith significant characteristic. In this study, de-esterification of apple pectin with acid treatmentresu...

Ultrasound Assisted In Situ Esterification of Rubber Seeds Oil for Biodiesel Production (RESEARCH NOTE)

Since the conventional esterification method requires longer processing time and obtain low yield, the intensification of this process is still interesting subject to be investigated. To reduce the oil extraction cost from seeds which almost 70% of total processing cost, in situ esterification has been recently introduced. The objective of study was to produce biodiesel from rubber seeds oil th...

Studies on the Enzymes of Pneumococcus

1. Pneumococci contain an intracellular enzyme of marked lipolytic activity as measured by the acid liberated by its action on tributyrin. 2. Enzyme-containing solutions may be prepared by dissolving pneumococci in bile, or by extraction by other means. 3. The optimum reaction for maximum activity of the endolipase is about pH 7.8, which coincides with the optimum hydrogen ion concentration for...

Batch Kinetics and Modeling of Alkaline Protease Production by Isolated Bacillus sp. (RESEARCH NOTE)

The aim of this study was the use of fish waste hydrolysate (FWH) as a substrate for alkaline protease production using isolated Bacillus sp. in a batch system. Then the fermentation kinetics of enzyme production was studied. The results show that with the addition of FWH to the fermentation medium with a final concentration of 4% (optimal concentration), alkaline protease value reached a maxim...
